"Elizabeth Bastard December 6." - under heading '1759'* I think this was probably the burial of this Elizabeth, though I am not certain. *
There is no note in the PR to show any relationship or whether she was a child. I don't think the absence of a note indicates she was not - in all of the burials in St George's in 1759 not one is shown as a child or has a parent or husband named, nor in the 1758 entries on the same page; it seems almost certain that some of 16 burials will have been of children.
* It is, though, in the parish where James' was having children buried around this time appears to have been.
* At the time James' was having children there were only two other Bastard couples having children, one of which was his brother Sampson and his wife, so it looks like there were not a lot of Bastards in Exeter then.
* I think Elizabeth would have been likely to stay in Exeter (the cities were where people moved to when they couldn't find work locally, hence I think she is unlikely to move out but it is not impossible). She therefore probably married and / or died there. The only marriage I have found that could possibly be hers was in St Sidwell's on 4 Sep 1780 to a James Meloney, and looking for the burials of the couple they would appear to be the James buried St. Paul's Exeter in 1823 and Elizabeth buried in the same parish in Apr 1824. Elizabeth was shown as aged 74, implying she was born about 1749, some years before this Elizabeth, but the informant could have been mistaken (especially as James appears to have been born c. 1754).

The marriage is listed after those in 1753, and being January I assume it was in 1754. New registers were used from 1754, but perhaps the church didn't have one this early in the year. Alternatively, the entry may have been forgotten and squeezed in at the end of 1753 when it was actually at the start, but I think this is less likely. It is unfortunate that whilst all the entries for 1753 include the year this one does not!
Is the marriage of this James and are the children those of this James and Ann? I think the evidence is strong that they are, given the following:
* Bastard was not the commonest of surnames, and it would seem the most likely James to marry there was the one baptised there, although another could have moved to the Ciy.
* James was a younger son in a gentry family, and the baptism I believe to be that of his wife-to-be shows her family were gentry.
* The names given to the children are probably the most convincing evidence - Rose and Sampson, not the commonest of names, Rose in particular being unusual, but the name of a sister and an aunt of James, as well as being the maiden name of his grandmother, while Sampson was the name of his father and a brother.
* While I am not at the time of writing sure of what became of James, his brother William mentioned him (living) in a will written in 1766 so he was clearly still living at the time of the marriage.
